![]() ![]() ![]() Suffice it to say, my first time trying a ready-to-go margarita cocktail in the form of the Kokomo Margarita Ice Pops was not the experience I had hoped for. It also robs you of the opportunity to decide just how strong or sweet you want your margarita to be. And while they are still cheaper than buying your own tequila and margarita staples, their basic taste and short freezer life don’t make them a viable option. At that point they are essentially watered down lemonade.Īt $12 a box, I hoped the Kokomo Margarita Ice Pops would be a cheaper alternative to making my own margaritas. I found it best to consume the freeze pops no later than two days after putting them in the freezer, whereas afterwards they tended to lose their smooth drinking texture and become rock solid. There were only small hints of lime and the alcohol was barely detectable. The taste of the freeze pops was lacklustre at best. Enjoying the margarita pops freezy style proved to be quite the undertaking, especially after the freeze pops had stayed in my freezer for a few days. The packaging says you can either have the freeze pops freezy-style or plop them into a cup. ![]() First off, Kokomo’s freeze pops are incredibly difficult to place in a glass. Unfortunately, my experience with the freeze pops was not as positive. The Kokomo brand also has mojito and cosmo varieties of their alcoholic freeze pops. The box made it look like everything you would expect from a regular margarita, without the need to go through the hassle of mixing it yourself. Here was a product which comes in a convenient box of six ready-to-consume margarita cocktails, but frozen. But on my last trip to my local Sobey’s liquor store I saw the Kokomo Margarita Ice Pops and was intrigued. Making a margarita at home is part of the fun - mixing the tequila, shaved ice, lime, and brimming it with salt. The delicious mix of tang and sweet with a side of salt makes for the perfect summer drink. While I’m a strong proponent of crisp craft beer or a neat Cuba libre, you can never go wrong with a classic margarita. With the heat rising, I wanted an easy way to enjoy a classic refreshing iced margarita cocktail. It is this ship, with substantial changes, that was the baseline model for the Valkyrie-class fighters in the Activision PlayStation game Star Trek: Invasion.Star Trek Online Tips for New Starship Captains by Trace_Thorne The 24- meter-long intermediate step between shuttle and runabout, carrying a crew of up to four, was armed with four type 5 phasers and three micro-torpedo launchers. This vessel is listed in role playing games, such as those from Last Unicorn Games and Decipher, as a Talon-class ship. Star Trek: Armada and Star Trek: Armada II give this vessel the classification of Venture-class, a starship with a sensors that can be modified to detect cloaked enemies. Starfleet used this class for research missions where they could be used for gathering scientific data and give assistance to survey teams in planetary and cultural surveys. 658))įor further information on the studio model, see Federation mission scoutship model.Īccording to Issue #80 of the Star Trek: The Official Starships Collection, this class was approximately 24 meters in length, had a maximum speed of warp factor 5, and a crew complement of one pilot and could carry 5 to 10 passengers. The cockpit of this scout ship, listed as "Starfleet scout ship", was a redress of the Class 2 shuttle Cochrane built for Star Trek: Voyager. ( Star Trek: Insurrection)īecause Michael Piller wanted the scene involving the scoutship to be similar to a World War I and II dogfight – in which both pilots were in open cockpits and could see each other – John Eaves created a vast windshield at the front of the scoutship. The vessel was accessible through its docking hatch on the ventral hull. It was slightly larger in size than the Type 11 shuttlecraft and carried dual-mounted phaser banks and photon torpedoes. ![]() The scout was a single- pilot starship with a forward mounted cockpit separated from the rest of the vessel. ![]() ![]() This will make a new window, holding the table you wanted. With the settings fixed, you can hit the “Create Table” button at the top of the main workspace. To do this, double click on that row, and select the statistic you want to include.Īnd you should now have something that looks like this: For example, I find it much more informative to have the singlets show total count, rather than Freq of parent. Some of those statistics aren’t what we’re looking for. Once in this window, select the populations / statistics you want to include from the main workspace, and drag it into the table editor, so you have something that looks like this. Lastly, the easiest way to output this data is to hit the Table Editor button near the top of the screen to open up a new window. Of course, a scatterplot needs a second axis, so I just used mCherry fluorescence (or the lack of it, since these were just normal 293T cells), captured by the YL2-A detector.Īnd of course copy that to the group as well (you should know how to do this by now). While this can be done in histogram format, I generally also do this with a scatterplot, since it allows me to see even small numbers of events (which would be smashed against the bottom of the plot if it were a smoothed histogram). In this case, it’s GFP positivity, captured by the BL1-A detector. Next is actually setting up the analysis for the response variable we were looking to measure. Then only gate the cells directly on the diagonal, thus excluding those that have more FSC-A relative to FSC-H. To do this, make a scatterplot where FSC-A is on the X-axis, and FSC-H is on the Y-axis. Here is the best description of doublet gating I’ve seen to date. So the live cell population will already be enriched for singlets, but having a second “doublet gating” step will make it that much more pure. Once you do that, the population should now be in bold, with the same text color as the group name. Thus, right click on the “Live” population in the workspace and hit “Copy to Group”. Once you have made that gate, you’ll want to keep it constant between samples. Since we’re mostly using HEK cells, that means that main thing we will be doing in this screen is gating for the population of cells while excluding debris (small FSC-A but high SSC-A). Set it so forward scatter (FSC-A) is on the X-axis, and side scatter (SSC-A) is on the Y-axis. By default, a scatterplot should show up. the first sample), and double clicking on it. Start by choosing a representative sample (eg. ![]() ![]() Now to actually start analyzing the flow data. Now that the group is made, I select it, and then drag the new sample files into it, like below: ![]() Thus, I hit the “Create Group” button and type in the name of the group I’ll be analyzing. Before you start dragging in samples, I find it useful to make a group for the specific set of samples you may want to analyze. With that out of the way, now you can perform your analysis. Thus, click on the “Preferences” button:Īnd finally once in that final window, change the “Decimal Precision” value to something like 8. This can be prohibitively uninformative if you have very low percentages that you’re trying to accurate quantitate. Once logged in, you’ll be starting with a blank analysis workspace, as below.īefore I forget, an annoying default setting of FlowJo is that it only lists two decimal points in most of its values. ![]() I’m obviously not going to share my login and password here ask someone in the lab or consult the lab google docs. Obviously, if you don’t have FlowJo yet, then download it from the website. Thus, here’s a short set of instructions for using it to do a basic process, such as determining what percentage of live cells are also GFP positive. While I’ve been using FlowJo for a long time, I realize it isn’t super intuitive and new people to the lab may first struggle in using it. Inevitably, what ends up being the most practical tool for analysis of low cytometry data is FlowJo. We do a lot of flow cytometry in the lab. ![]() ![]() ![]() ![]() A bit over dramatic, but then that's to be expected. What would have happened if they just killed the herders? What would have happened if they tied them up, went a bit down, risked a sniper shot at the enemy commander, then just ran? What would have happened if the Pashtuni would have ignored the wounded American or would have killed the Taliban scout force when they came to them? How would the mission have gone if the four guys would have known from the get go that they would be completely alone, with no support or hope for extraction? Overall, a very emotional movie, two hours long, that shows more a general type of heroism than one with a specific purpose. Director Peter Berg took on an important task when he brought a heart-wrenching true war story to life. As they confront unthinkable odds, the SEALs must find reserves of strength and resilience to fight to the finish. I liked a lot about the movie how it made you think long after it was over. Four Navy SEALs on a covert mission to neutralize a high-level Taliban operative must make an impossible moral decision in the mountains of Afghanistan that leads them into an enemy ambush. The story, while showing the preparation, courage and resilience of four soldiers in enemy territory, also showed other things, like the logistical blunders that lead to stupid deaths, over-reliance on technology that doesn't really work as you expect and how choices have consequences on the ground that are beyond the ability of normal courts to understand, whether looking from the legal or moral angle. The actors also play well, although I like Mark Wahlberg in almost everything he does. You will witness two hours of low tech war in all of its horror and stupidity. On the other hand, the fights were realistic, the subject based on real events and, outside the pathetism described above, I did not detect a bias towards one side or the other. People in the field try to carry out their mission and survive, while their deaths become political and mediatic material. Also showing pictures of dead soldiers with their families with a pathetic American remake of Bowie's Heroes singing in the background at the end of the movie just fueled more rage. Having all Americans die in slow motion while scores of Taliban died instantly and kind of stupidly didn't help with the empathy. I wanted the characters to succeed, to survive, but I could not ignore the fact that they were soldiers being there only to kill an enemy commander. Raytheon should be annoyed that a movie about a mission failed primarily because of communication issues showed their red flashy brand on the comms equipment. For myself, I felt mostly rage against a botched mission in an ineffective war. Now, what you will feel will no doubt be up to you. I assure you you will not get bored watching it. ![]() , free movie online xmovies8, xmovies8.si, xmoives8, x8movies com, xmovies8,tv,, xmovies8 tv.If there is one thing that this film will accomplish is to make you feel something. Uwais has also become one of the biggest martial arts stars in the world, continuing to wow audiences in movies like Beyond Skyline or The Night Comes For Us, along with the Netflix series Wu Assassins and its upcoming movie sequel, Fistful of Vengeance. Though The Raid 3 never happened, both Evans and Uwais have gone on to great careers, with Evans directing the aforementioned Apostle and creating TV series Gangs of London. Evans also seemed intent to hold true to Rama's minimal involvement in the third movie, with The Raid 2 ending on a weary Rama declaring " No, I'm done" when approached by Goto. The American remake of The Raid remains in development , though a parting of ways between rights owners XYZ Films from director Joe Carnahan and star Frank Grillo now makes it an unofficial one on the latter pair's part titled Zeno. From the sound of it, The Raid 3 was going to take quite a left turn from the end of The Raid 2. The film would also have been set mostly in the jungles of Indonesia as the Gotos battle their Yakuza bosses. Instead, the film would have focused more on the Goto crime family coming into conflict with their Yakuza superiors. Evans explained in a 2016 interview with Impact Online (via AICN), " Whether it was a conscious decision or not, moving back to UK felt like a closing chapter on that franchise - we ended the story pretty neatly (I feel) in Part 2." Evans also reiterated this point while promoting Apostle, stating in an interview with Slash Film, " What we did with The Raid 2, we kinda close that off nicely, so it didn’t really appeal to me to jump back into that world again".Įvans has also stated Uwais' character Rama would've had a minor role in The Raid 3 compared to its predecessors. ![]() The likes of Chris and Liam Hemsworth and Frank Grillo have all been. A combination of the ending of The Raid 2 and Evans' return to the UK from Indonesia led to him moving on from The Raid 3. An American remake of The Raid has been gestating for almost a decade, but has never quite got off the ground until now. RELATED: Warrior's Best Fight Borrows Heavily From The RaidĮvans had already wanted to take a break from action moviemaking in the interim between The Raid 2 and The Raid 3, with his next film being the 2018 Netflix horror Apostle. Though Evans conceived of The Raid franchise as a trilogy, The Raid 3 ultimately never happened, with a combination of factors explaining why. Its 2014 sequel The Raid 2 continued to turn the volume up with a more complex crime family plot and even wilder action. Following a police raid of a Jakarta apartment complex filled with killers and psychopaths, The Raid combined the Silat-based action scenes of Merantau with the tone of a survival horror game and became an instant action classic. ![]() ![]() ![]() One can always use the boots without the insoles and they are designed for sockless barefoot use, although I prefer socks inside boots. Total stack height is about 11MM, but 3MM less without the softer insoles. The total thickness of the outsole is good for protection, yet connection. The treads are significant for a minimalist boot. I suggested to people they use a match or lighter to melt that away if it bothered them and that works, but I don’t think that will be an issue with this new model.
